LESSON 21: P141 Peptide: Male and Female Libido

PT-141, also known as bremelanotide, is a synthetic peptide derived from α-MSH (alpha-melanocyte-stimulating hormone).

It is primarily studied and used for:

* Low libido
* Sexual dysfunction
* Arousal disorders

Unlike traditional ED drugs, PT-141 is not vascular-based—it works in the brain.

⚙️ Mechanism of Action (Why It’s Different)

PT-141 activates melanocortin receptors (MC3R, MC4R) in the brain—especially in the hypothalamus, the control center for sexual behavior.

🧠 What this means:

* It increases desire first, not just blood flow
* It stimulates neural pathways of arousal
* It can trigger both psychological + physical sexual response

👉 Think of it like:

“Turning up the brain’s arousal switch—not just fixing the plumbing.”

🔥 Effects on Arousal & Libido

Key Effects:

* Increased sexual desire (libido)
* Enhanced arousal response
* Improved erectile response (men)
* Increased sexual satisfaction (women)

Clinical and preclinical data show:

* Rapid onset (often within ~30–60 minutes)
* Improved arousal and satisfaction scores in women with HSDD
* Induced erections in animal and human studies

⚖️ Clinical Use

PT-141 is FDA-approved for:

* Hypoactive Sexual Desire Disorder (HSDD) in premenopausal women

⚠️ Off-label research/interest includes:

* Male erectile dysfunction
* Performance anxiety-related dysfunction
* Libido enhancement

💉 Administration (Educational Only)

Common delivery methods:

* Subcutaneous injection
* Nasal spray

Typical research protocol:

* Taken ~45 minutes before activity

⚠️ Side Effects & Risks

Common:

* Nausea
* Flushing
* Headache
* Increased blood pressure

Important considerations:

* Acts on central nervous system
* May affect cardiovascular parameters
* Not suitable for uncontrolled hypertension
